【発表概要】
今回のサイエンスカフェでは、リボソーム病の話題について提供したいと思います。
RMRPはRNase MRPを形成するnon-coding RNAで、RMRPの変異は骨格異常とT細胞活性化障害を特徴とする軟骨毛髪低形成(CHH)を引き起こすことが知られています。筆者らは、CHHでみられる変異が細胞質リボソームの形成異常に関わることを見出し、CHHがリボソーム病であることを示しました。

RMRP encodes a non-coding RNA forming the core of the RNase MRP ribonucleoprotein complex. Mutations cause Cartilage Hair Hypoplasia (CHH), characterized by skeletal abnormalities and impaired T cell activation. Human cells engineered with the most common CHH mutation (70AG in RMRP) show specifically impaired pre-rRNA processing, resulting in reduced mature rRNA and a reduced ratio of cytosolic to mitochondrial ribosomes. These results indicate that CHH is a ribosomopathy.
