“In vitro model of FTLD-Tau shows impaired axon initial segment (AIS) plasticity and hyperexcitability”

タウ病理を伴う前頭側頭葉認知症(FTLD)は神経回路の異常興奮性に関連している。 Sohnらは タウ変異を有するヒトiPSC由来神経が、AISの細胞骨格異常を原因とする軸索起始部(AIS)における活動依存性可塑性の消失および神経活動の恒常性異常を示し、結果として神経ネットワーク機能の調節不全が引き起こされていることを報告した。本発表では、FTLDにおけるAISの重要性について、実験の方法論にも着目しながら議論したい。
Frontotemporal lobar dementia (FTLD) with tau pathology is associated with aberrant hyperexcitability of neuronal networks. In human iPSC-derived neurons, Sohn et al. demonstrates that FTLD-causing tau mutation abolishes activity-dependent plasticity of the AIS and impairs homeostasis of neuronal activity via impacting AIS cytoskeleton, resulting in dysregulation of neuronal network function. In this science cafe, I would like to discuss about not only the importance of AIS in FTLD but also the experimental methodology.
