細胞内のタンパク分解機構については、ユビキチン-プロテアソーム系及びリソソーム系など解明が進んでいますが、細胞外の異常タンパク質を除去する分子機構はまだよく解明されていません。本研究グループは以前の研究で、細胞外シャペロンであるクラステリンが細胞外変性タンパク質をリソソーム分解へ導くことを明らかにしています。今回は、クラステリン以外に、α2マクログロブリンがその作用を有することを報告しています。
【Summary】
The mechanisms of intracellular proteostasis, including the ubiquitin-proteasome system and lysosomal system, have been known. However, the molecular mechanisms that remove extracellular misfolded proteins are still poorly understood. In a previous study, this research group showed that the extracellular chaperone prorein, clusterin, acts as a substrate for extracellular misfolded proteins to lysosomal degradation. In the present study, they indicated that α2-macroglobulin, in addition to clusterin, acts as another clearance factor.
