Senolysis by glutaminolysis inhibition ameliorates various age-associated disorders

今回のサイエンスカフェでは、「老化細胞の生存戦略」と「その阻害による若返り」について東京大学医科学研究所の興味深い発見を紹介したいと思います。
老化細胞ではリソソームに損傷が生じることで細胞内pHが低下しますが、これを回避するためアンモニアの産生を亢進する働きが細胞内で活性化します。逆にそれを阻害することで老化細胞を除去しアンチエイジング効果が得られるという報告です。本発見に際してキーテクノロジーとなった「大規模定量プロテオミクス:iMPAQT」についても紹介致します。
【Summary】
In the next Science Café, I would like to introduce an interesting discovery made by the Institute of Medical Science, University of Tokyo, regarding the survival strategy of senescent cells and rejuvenation through their inhibition.
In senescent cells, damage to lysosomes causes a decrease in intracellular pH, and to avoid this, the production of ammonia is activated in the cells. Inhibition of GLS1, a key protein in ammonia production, can eliminate senescent cells and produce anti-aging effects. In this presentation, we will also introduce the key technology behind this discovery, i.e., large-scale quantitative proteomics: iMPAQT.
