【発表概要】
Gタンパク質共役型レセプター(G-Protein Coupled Receptor:GPCR)は、創薬標的の最大のファミリーの一つです。GPCRのシグナル伝達には、Gタンパク及びβアレスチンの2経路が知られています。最近では、副作用軽減を目指して、選択的な活性化を示すbiased ligandの開発が行われています。しかしながら、βアレスチンの活性化メカニズムについては、完全には解明されていません。著者らは、1分子FRET法を活用して、活性化されたβアレスチンのC tailの挙動を明らかにしました。
【Summary】
G-Protein Coupled Receptors(GPCRs)are one of the largest family of drug targets. It has been reported that there are two pathway of the GPCR signaling : G-protein and β-arrestin. Recently, the biased ligands showing selective activation are developed in order to reduce side effects. However, the activation mechanism of β-arrestin is not fully understood yet. In this paper, the authors revealed the motions of C-tail of the activated β-arrestin by using single-molecule FRET(smFRET).
