【発表概要】
SNIPERやPROTACsのタンパク分解誘導技術(targeted protein degradation:TPD)は、標的タンパクのリガンドとユビキチンリガーゼのリガンドをリンカーで連結することで、標的タンパクへ強制的にユビキチンを付加させ分解誘導することから、新たな創薬手法として注目されている。しかしながら、これらの技術は、主に細胞質内のタンパクを標的とした手法であり、細胞膜上のタンパクや細胞外タンパクは対象外である。著者らは、細胞膜上に存在し、リソソームへシャトリングするcation-independent mannose-6-phosphate receptor(CI-M6PR)のリガンドと標的タンパクのリガンドを連結したlysosome-targeting chimaeras(LYTACs)を開発し、細胞膜上或いは細胞外の標的タンパクに対してもリソソーム経路で分解可能なことを示した。また、CI-M6PRの代わりに肝臓特異的に発現しているシャトリングタンパク(ASGPR:asialoglycoprotein receptor)を利用することで、肝臓細胞で特異的に標的タンパクを分解誘導する実験データも示している。そのため、本技術は、TPDの標的タンパクの可能性を広げ、創薬への応用が期待される。
【Summary】
SNIPER and PROTACs are chimeric compounds connected between a target protein ligand and a ubiquitin ligase ligand by linker. These techniques enforce to transfer ubiquitin proteins to targeted protein, and degrade by ubiquitin proteasome system. Targeted protein degradation (TPD) has attracted a lot of attention as a new drug modality. However, these techniques are mainly targeted to the intracellular proteins, except for extracellular and membrane-associated proteins. Authors established lysosome-targeting chimaeras (LYTACs) which connected between the ligand for cation-independent mannose-6-phosphate receptor (CI-M6PR) expressed on plasma membrane and shuttled to lysosome, and the ligand for targeted protein. Authors indicated that LYTACs degraded extracellular and membrane-associated proteins by the lysosome pathway. Moreover, authors used the ligand for asialoglycoprotein receptor (ASGPR) which express specifically in liver cells instead of CI-M6PR, and reported downregulation of a membrane protein in liver cells. Therefore, LYTACs technique would expand the possibility of targeted proteins for TPD, be expected to apply for drug discovery.
