ロンドン王立大学のグループから発表された本論文は、マウスの脳内でヒトiPS由来の神経のシナプス形成や刈り込みを長期間観察できることを示した論文であり、昨年の11月16日号のScience誌に掲載されました。特筆すべきは、トリソミーを持つダウン症の神経細胞は、軸索に沿って形成されるスパインとよばれるシナプスが、健常人由来ニューロンと比較し、安定で刈り込みされにくいため、スパインの密度が増えていることを発見した一方、ニューロンの活動は強く低下していることを研究グループは明らかにしました。
Xenotransplantation(ヒト細胞キメラマウス)の研究は複数のグループで行われていますが、このサイエンス誌の論文に加え、bioRxiv誌に現在掲載されているベルギーLibre de Bruxelles大学の研究グループのプレプリントの論文(https://www.biorxiv.org/content/10.1101/626218v1) も手短に紹介し、このXenotransplantationが持つ将来性を概説させていただく予定です。
“In vivo modeling of human neuron dynamics and Down syndrome”
This paper, published by a group at the Royal University of London, shows long-term observation of synapse formation and pruning of human iPS-derived neuron in the brain of mice. It should be noted that Down's syndrome neurons with trisomy have a higher density of spines because synapses in spines formed along axons are more stable and less likely to be clipped compared to neurons from healthy individuals. The research group revealed that it caused strongly reduction of neuronal activity. Research on Xenotransplantation (human cell chimeric mice) is being conducted by several groups in addition to this science journal paper. A preprint paper by a research group at the University of Libre de Bruxelles in Belgium, currently published in bioRxiv (https://www.biorxiv.org/content/10.1101/626218v1) will also be briefly introduced to outline the future of Xenotransplantation.
