Xist ribonucleoproteins promote female sex-biased autoimmunity

【発表概要】
Xist long noncoding RNA(lncRNA)は、2本のX染色体のうち1本をランダムに丸ごと不活性化することで、X染色体コード遺伝子の発現量を補正するために女性でのみ発現する。本研究では、Xist ribonucleoprotein複合体が、多数の自己抗原性成分を含んでおり、性差のある自己免疫の重要なドライバーであることが示された。ヒト自己免疫疾患患者では、XIST RNPの複数の構成要素に対して自己抗体を示しており、性特異的なlncRNAがユビキタスRNP構成因子とともに性差による自己免疫を駆動していることが明らかとなった。
【Summary】
Xist long noncoding RNAs (lncRNAs) are expressed only in females to compensate for the expression of X chromosome coding genes by randomly inactivating one of the two X chromosomes in its entirety. The present study demonstrates that the Xist ribonucleoprotein complex contains numerous autoantigenic components and is an important driver of sex-biased autoimmunity. Patients with human autoimmune diseases exhibit autoantibodies to multiple components of the Xist RNP, indicating that sex-specific lncRNAs, together with ubiquitous RNP components, drive sex-biased autoimmunity.
