【発表概要】
SARS-CoV-2とSARS-CoVの間の抗原性の違いに関する情報はワクチン開発に重要な意味を持つ。しかし現時点ではSARS-CoV-2の感染により誘発された抗体反応に関するデータは少ない。特に免疫学的課題の1つとしてのSARS-CoV-2とSARS-CoV間の抗原性の違いに関する情報が不足している。
SARS-CoV-2またはSARS-CoVに感染した患者の血漿と、これらの両者に感染または免疫したマウスから得られた血漿を解析した結果、スパイクタンパク質に結合する抗体の交差反応は一般的であったが、交差中和はまれであることが示唆された。
この事実はスパイク内中で保存されたエピトープに対する非中和抗体応答の存在を示しており、この反応が抗体依存性病態増加につながるかについてより詳細な研究が必要であることを示している。
【Summary】
There is currently a lack of knowledge in the antibody response elicited from SARS-CoV-2 infection. One major immunological question is concerning the antigenic differences between SARS-CoV-2 and SARS-CoV. By using plasma from patients infected by SARS-CoV-2 or SARS-CoV, and plasma obtained from infected or immunized mice. These results show that while cross-reactivity in antibody binding to the spike protein is common, cross-neutralization of the live viruses is rare, indicating the presence of non-neutralizing antibody response to conserved epitopes in the spike. Whether these non-neutralizing antibody responses will lead to antibody-dependent disease enhancement needs to be addressed in the future. Overall, this study not only addresses a fundamental question regarding the antigenicity differences between SARS-CoV-2 and SARS-CoV, but also has important implications in vaccine development.
