要旨:
筆者らはナノスケールでその構造を自由に設計できるDNA origamiとDNA アプタマーを組み合わせることで、分子応答性ドラッグ放出キャリアー “DNA nanorobot”を構築した。本キャリアーにはトロンビンが搭載されており、がん細胞表面で過剰発現しているヌクレオリンと相互作用すると、鍵となっているDNAアプタマーが”開錠”される。担がんマウスに本キャリアーを静脈投与すると腫瘍部位のみで血管内血液凝固を引き起こし、腫瘍の壊死および増殖阻害をもたらすことが示されている。

Summary:
The authors constructed a molecule-responsive drug-release carrier “DNA nanorobot” by combining DNA origami and DNA aptamer, whose structures can be freely designed at the nanoscale. The carrier is loaded with thrombin and interacts with nucleolin that is overexpressed on the surface of cancer cells, which unlocks the key DNA aptamer. Intravenous administration of this carrier to tumor-bearing mice has been shown to cause intravascular blood coagulation only at the tumor site, leading to tumor necrosis and growth inhibition.
